A primary character is an employee at Planet Express, with the notable exception of Scruffy. They are primary characters, as they appear in almost all episodes, comics and films.

Fry, Leela and Bender are considered major primary characters, or main characters, because they appear in every episode, comic or film, have by far more A-plots revolving around them. The Professor, Hermes, Zoidberg and Amy are considered minor primary characters, becuase they appear in almost every episode, but have considerably less A-plots revolving around them when compared with the major primary characters.
